### Setting corresponding values for blur.mu and blur.sigma

Hi,

In the multispot5.cfg file, we were supplied with three sets of values for blur.mu and blur.sigma according to the pixel size (100, 160 and 79nm per pixel). For pixel sizes not listed here (from custom setups), are we Okay by using values from the pixel size closest to the actual value? Alternatively, how can we determine these values based on the pixel size?

It's probably OK to use the nearest if it's not too far out.

Alternatively, how can we determine these values based on the pixel size?

Assuming you have the FWHM in nm and the pixel size in nm, use the equation:

$$\text{blur.mu} = \ln\left(\frac{\frac{\text{FWHM}}{\text{pixel size in nm}}}{2 * \sqrt{2 * \ln 2}}\right) + (\text{blur.sigma})^2$$

We always use blur.sigma=0.1
